Function:
Ezetimibe is a new class of selective cholesterol absorption inhibitors that inhibits the delivery of small intestine to the diet and bile to the intestine by binding to the small vesicle membrane protein (relative molecular mass 145X103) in the small intestine brush border membrane. The absorption of cholesterol lowers the cholesterol content in serum and liver. Unlike bile acid chelating agents, ezetimibe does not affect the absorption of cholesterol esters, other terpenoids (such as taurocholic acid), triacylglycerols, and fat-soluble vitamins. Its pharmacological action is independent of the inhibition of acetyl-CoA cholesterol acetyltransferase (ACAT) and the expression of LDL receptor (scavenger receptor). After being absorbed, ezetimibe is bound to glucuronic acid in the liver and then circulated through the liver and intestine, and is almost exclusively localized to small intestinal mucosal cells.
